Wockhardt Share Price Surges Over 8%; Secures CDSCO Nod for Antibiotic Zaynich in India

Wockhardt has advanced its anti-infective portfolio with ZAYNICH, a first-in-class antibiotic developed for adults with complicated urinary tract infections and associated Gram-negative bloodstream infections.  

The therapy is supported by positive clinical study outcomes and strengthens the company's focus on tackling antimicrobial resistance. 

Late-Stage Study Shows Higher Success Rates 

The company's ENHANCE-1 Phase 3 trial evaluated ZAYNICH against meropenem in patients with complicated urinary tract infections, including pyelonephritis. The study measured both clinical cure and microbiological eradication. 

ZAYNICH achieved the primary endpoint in 89% of patients, with 250 out of 281 participants responding successfully. Meropenem recorded a success rate of 68.4%, with 93 out of 136 patients meeting the same endpoint. The difference of 20.6% points favoured ZAYNICH. 

The results were assessed around 10 days after completion of treatment. Among patients who also had Gram-negative bacteremia at baseline, ZAYNICH delivered an 89% response rate compared with 44% for meropenem. 

Backed By Multiple Clinical Studies 

Before entering Phase 3, the antibiotic completed nine Phase 1 studies and a Phase 2 study conducted across 15 leading tertiary-care hospitals in India. 

The Phase 2 study focused on meropenem-resistant Gram-negative infections and reported clinical efficacy of more than 97%. The trial included patients with hospital-acquired bacterial pneumonia, ventilator-associated bacterial pneumonia, bloodstream infections, complicated intra-abdominal infections and complicated urinary tract infections. 

The company believes the treatment can address carbapenem-resistant and extensively drug-resistant Gram-negative infections, areas where existing therapies can face efficacy and safety challenges.  

ZAYNICH has also been designed to tackle metallo-β-lactamase-mediated resistance, a significant resistance mechanism seen in India. 

Product Profile and Pipeline 

ZAYNICH combines Zidebactam and Cefepime in an injectable formulation. The 2 compounds target different penicillin-binding proteins, helping eliminate Gram-negative bacteria through a dual-action approach. 

The therapy has also demonstrated encouraging outcomes in 85 compassionate-use cases involving extensively drug-resistant Gram-negative infections across India, the United States, Malaysia and France. 

Wockhardt currently has 6 antibiotics in its pipeline, split equally between Gram-negative and Gram-positive infection treatments. The company employs around 3,200 people across global operations, with international markets contributing 78% of overall revenue.
